NO TYPO.

Everytime i open my folder and see this:

i think it's a shorthand or typo for "genome" and think it's this:

but i was in my right mind when i named the folder. it's actually this:

last night i went to eat at genki with le boyfriend, uncle ken, jet tora and a new friend, theresa! so nice to end the week with good company. theresa is a nursie who works nightshift (with babies!) so we swapped some stories. :) we went to starbuckss after the food, and then to walmart to check out the bargain bin dvds.

le boyfriend bought The Fourth Kind

i thought it was scarier than Paranormal Activity 2. after the initial shock, Paranormal seemed kind of fake to me. i lost interest at the being dragged down the stairs part. :/ but in this movie, there are these freaky video recordings of hypnosis that gives me the creeps! scary movies are good fun times. heeheee. and did i end up staying up all night looking for owls outside my window?

i slept like a log.

when you're tired, you're tired. :)

as for my videos, i picked up The Zombie Diaries ($9) and Batman Forever ($5?!!!) so will probably watch some zombies tonight. :)
i really want to find the movie The Signal. one of my favorite pseudo-zombie/brainwash movies. another movie i want to see is GATTACA (named after EcoRI!) but i want to rent it before buying.

that's all for now, i guess.
tomorrow is halloween! i haven't settled on a costume yet and i work from 0600-1430 so time is running out for me...

random stuffs

got the tentative schedule for november and december. it's official! i'm going to night shift. :) just a couple weeks more of blood bank and chemistry (yes, i know i've been saying this forever but it's finally feeling so real!) and then the next chapter of my life begins. it's awesome how everything seems to be falling into place now. but the most interesting kismet is: thwo of the other night shifters are named kori and cory. how amazing and coincidental is that?! it was meant to be! adding a codi is going to make things confusing maybe but i guess that's part of the fun. XD

anyway, at work today we were totally kicking butt in blood bank. one person was out today so i started the morning by myself... with a type and screen! (plus morning reports, QC, and stuffs) luckily, debbie came to save meeee after a bit. then i worked with myra for the bulk of the day. i guess there are a lot of surgeries coming up so there were a lot of type and screens. then both myra and i had antibodies at the same time! i had a previously identified antibody but it was over a month ago PLUS the patient had a warm autoantibody so it masks everything with a positive. :p had to cancel the procedure and send the samples out to BBH for a work up. myra was not so lucky- she had to screen A LOT of units to find two M-negatives. A LOT of units. heeheee. in the mean time i was picking up some stats and ordering stuff from BBH so the day flew by. in a good way. after 1pm, things steadied to a lighter pace so i hope it stays that way for eves and nights.

yesterday, i went to ER to draw. i haven't done it in a while! and of course, dramas. one patient was a super hard stick! the doctor was mumbling about peripheral vascular disease. they let me sweat for about 30 minutes, trying to find some veins... any veins... and then they said never mind and drew from the jugular for me. *roll eyes* thanks guys. another phleb (lindalove!) tried to draw the same patient before me and was unsuccessful. blah, i wish the doctors would pity us sooner! the second patient i saw was having breathing problems. i got coughed on a few times. :/ but i got the blood. and then as i was leaving the patient revealed they had TB 30 years ago. i was like "waaaaaaaaaaaaaat.... " À_____Á i'm it's all cleared up now (modern medicine FTW) and the chest xray was promising. but i was feeling really paranoid. ahhhhg. hate when that happens.

tonight is the finale of project runway! i am so excited. i can't wait to see the looks. more than i care who is the winner or how it was made... i just want to see the fashions! i love to find new and unique outfits. plus... gotta see the hawaii boy represent!! (but i have my money on mondo as of now.)

the white bunny that lives on our street (wild) got hit by a car last thursday. T_____T
i am so sad! i wanted that bunny to be mine!

551414-R1-05-6

now they keep the black one (my favoritest one) caged up so i don't see her anymore either.
soooo saaaaaaaaaaad. TT_________TT

ever since i started eating the spicy instant noodles, the korean one from don qui called "hwa ramyun" (lol the package says "with soy peptide!" whatever the hell that is) everything gets the volume turned waaaay down. i put so much siracha in my wonton mein tonight. heeheheee. my dad made the won tons, by the way. they are tha bomb!! they sing of deliciousnesss. plus he folds them nice and tidy! i'm so full now... ate too many tons for dinner. ^o^

how to: Lift and mount the emulsion of polaroids

this process makes your polas stable for eternity. :D
(i love the little "yes!" fist pump after the successful separation.)
it is so thinnn! it is reminding me of making strussel dough. :P
will i buy a lift kit? maybe not. likely not. but can i resist the other films??? oh ho ho... my weakness! i am dreaming of the black and white UV. * u* i need to get my pioneer card before it's too late! but i've decided not to buy any more camera stuffs for a long time! have to save money, ahhhh...

in fact, i'm going on a shopping strike for a while! (U___U) but a girl can still dream~ aren't these boots cute?

madden boots

things i am NOT liking: ruched sleeves.

bad sleeves

no.
just... no. this is so fuggggly. it looks unnatural. why would you scrunch up your sleeves on your shoulder? elbow, yes. shoulder. noooooo.
exaggeration? maybe. but i can't stand this look. >:U

let's end on a happy note~

Pete Souza. *swoon*
his photography blows my mind sometimes.

P080910PS-0356

yes, i do follow the white house flickr in my rss feeds. the photos that mr. souza (and the other official photogs) take portray mr. obama as youthful, candid, and curious. it's interesting to see what goes on in the serious times as well as the "down time" of the president. (does the president ever really have down time? you can't exactly take a vacation from being president.) it makes the important subject very humanizing and sometimes humbling.

want to know what it's like to be an official white house photographer? Q&A session with pete souza on october 28th. more info here: white house flickr feature!

today is my first day as a solo blood banker (woohoo, signed off on my training! only coag and chem to go!) and guess whaaaaaaaaat!? i got an antibody! i mean, a patient with an antibody! a previously unidentified antibody! oh la la! it turned out to be a Lewis (Lea) so it was easy enough to find units.
i also got to do a plfa which turned out to be abnormal (patient taking aspirin?) so those took up a good chunk of the day. we also had some BioRad people peddling their wares and they brought cookies. ^o^

lately, i can't get enough silversun pickups. i've been listening to dream at tempo 119
nonstop. i fucking MELT to the song. it's so dreamy.

also good: little lover's so polite.

HEART IN A CAGE

:)

All our friends, they're laughing at us
All of those you loved you mistrust
Help me I'm just not quite myself
Look around there's no one else left
I went to the concert and I fought through the crowd
Guess I got too excited when I thought you were around
Oh he gets left,
left
left

And the heart beats in its cage

been tearing apart my room for months, trying to find my favorite pink bag with my favorite strokes patch. :'C

LESS THAN 16 OZ. OF COFFEE BEFORE 1200/PER DAY IS TORTURE

yesterday i went to the lab (the research lab) after work to see if my bacteria transformed. alas, disappointment. damn. everytime i do a transformation experiment it is a FAAAAAIL. even my vibrio transformation was a fail.

and my keratin transformation was a FAAAAAAAAAAIL.

Codi Keratin Cloning 100µl

i was even using a kit for the keratin clone project. ahgg!

clone kit

anyway, i am definitely going to repeat the experiment because i have no ideaaaa what went wrong.
pam was there working on protein gels too.

so so so... we worked together on vibrio

Vibrio fischeri glowing!

last year and she was also disappointed that it didn't work out. and we are going to repeat vibrio again!

breaking Vibrio (DNA precipitation)

i can't wait! we can do both transformations at the same time! i am looking forward to digging my competent cells out of the freezer again.

another weekend gone.

Things just look better in black and white, these days.

just shooting around

i can't believe summer is ending... ended. or, as ended as it gets in hawaii. but it issssss getting colder. i can tell! ergo, spent some time admiring the sky and the sunshine today with twinkies.

finally finished season three of dexter (thanks to the public library)!! such a complicated story but it still unfolded well, in the end. (not the way i wanted, but it was still a satisfactory ending.) i guess i better reserve season four and see what's next!

more random thoughts:
does anyone know if there is such a thing as licorice flavor lollipops and where can i get them? also, i am still looking for salty/salmiak licorice locally. i've only seen this online (i am leary of purchasing food online) and once, when a polish anthropologist gave me some (which she had gotten from poland). (i know i have blogged this plea before but i am dying to eat lakerol! T___T)

even more random:
i think horses are kind of gross looking and scary (horses are so fricken huge!) but i think llamas and alpacas are cute. how my brain made this decision (as i've seen neither up close) i have no idea. but i wouldn't mind hugging and having a pet llama or alpaca! i would also like to have sheep. they are so adorable. but bunnies and cats are probably a better size. of course my ultimate pet to have would be a bat! i've wanted a bat as long as i can remember. they are cute adn fluffy (not feathery) AND they fly! they have cute little fingers and little teeth. ^___^ chickens are also cute- but only if they look like the chickens in Castle Crashers.

chickennn!

and now, current musicssss:

All Heart- Roll the Credits (been listening to a lot of All Heart, recently. i need to see them live!)

ANKLE STRAP FLATS

daaaaaaaamn....
i've been looking for a pair of plain, black, ankle strap flats for a long time now (trying to recreate an outfit).
and i've found just what i'm looking for!
but i'm not going to buy them!
you see, i have [fallen in love with] Melissa plastic shoes that are plain, black, flocked and absolutely PERFECT (even sans ankle strap) so i am no longer hunting for ankle strap shoes.
in fact, i was so thrilled by the melissa shoes that i am always on the hunt for more! i have a clear-leopard melissa flat and orange-coral patterned flats (omg, found these at norstrom rack for $30! a deal unheard of!!).
anyway, i just figured there might be someone else out in the world looking for perfect black ankle strap shoes and might want to jump on these cuties (which happen to be a bargain and made of suede!).

Paper Pola Pals by =somnio-insania on deviantART
got up at 6 to tend to mom's finger- she smashed in the car door this morning.
after that i couldn't go back to sleep so i've been doing paper crafts in my room and drinking coffee and tweaking to whatever itunes has picked for me. i discovered that MONTH OF MAY by ARCADE FIRE sounds really great after UNTITLED by FINCH when played back to back seamless. the scream at the end of UNTITLED fades into the scratchy-ness of MONTH OF MAY. have a listen!

Fwd: Random cancer question

FYI, people.
i used to see this a lot in heme and was hoping a cancer expert/dr. of molecular biology could explain to me.
maybe some other time i'll find someone who can explain.

Codi,

One of the strange features of tumor cells is that they ball up, atleast in culture we see that and the theory is that the carbohydrates on the cell membrane are more mobile in the bilayer than regular cells.  Consequently they move around, stick to other things and allow for clumping.  The reason this happens is unknown.

As for the wbcs, I don't have an answer and could only throw speculative hypothesis out there, but I won't.

Sorry we didn't talk tonight, I'm pretty busy but if you peg me down I'll bring up the plasmid so you can transform sometime.

Hi mattT,
I was wondering if you could tell me why tumor cells "ball up" in body fluids.  Or why white cells cluster together in peripheral blood.
Just curious,

C.

* edit//
i got the pics by google image search for combinations of the following words: tumor cancer ball blood synovial body fluid csf differential blasts
i picked out out pics that look like what i see in differentials (basically, cells in wright stain)

the first pic is a good example of white cells getting too friendly- you never see white cells close together in normal diffs)
the next pic have blasts, but they aren't balling up.
the next is a crap load of macrophages... common in synovials but their bunching might be from the cytospin, not because they are malignant.
the last picture has blasts and the ones in between have blasts, synovial cells and other yummy fun stuff. (second to the last maybe from bone marrow? not sure.)
i didn't find any really good examples of balled up cells. when i go back to heme (i'm working blood bank now) i'll see if i can find some nice pics.

TWILIGHT GALAXY

first day back to work after day off is so haaaard~
blood banking today. was fun, nonetheless.
super busy since last night (i guess... i wasn't working last night *wink wink*) bleeders and antibodies and in patients. not to mention i needed a refresher in the computer system since i haven't blood banked since may.
i love pooling platelets and splitting units. something about watching the bags fill up with swirly liquid (i SWEAR if you look at platelets in the bags they swirl around. so magical!) is very satisfying. and then you put the giant stickers on A, rh POSITIVE (etc.) it's like saying "HA! i give you my stamp of approval!"

one thing i dislike about bb is doing refrigerator alarm checks in the morning. you have to call the security department first because testing the battery on the frigo sets off the alarms. it seems so trivial, i'd hate to bother the security (who probably don't even give a shiet about blood bank let alone their refrigerators) with such information. i'm sure they are busy with their own tasks of the morning like restraining combative patients and making sure no one runs off with someone else's babies.

got my flu shot today. all you people depending on herd immunity- YOU'RE WELCOME. ó____ó

going to start working on transforming E. coli with wintergreen plasmids. :)

besides that, i have nothing exciting to blog about.
except some crazy ass dream i had a few nights ago, but it's so complicated i can't really talk about it.

i guess it's pic spam time.
if anyone else wants to share their flickr pages with me, i'll make you a friend! i love browsing devart or flickr and reading magazines/blogs in my rss feed. it's like going on a gallery walk and reading lots of magazines every day. :)
